Abstract

The best antisecretory drugs for treating acid-related conditions are proton pump inhibitors (PPls). They 
are therefore often administered to patients who are taking other drugs at the same time. PPls may interact 
with other medications in a variety of ways. The two most significant ones are the competitive suppression 
of hepatic cytochrome P (CYP) 450 enzymes involved in drug metabolism and the modification of other 
medicines' absorption through pH changes in the stomach. Drug interactions may be more likely to occur 
in poor metabolizers who lack CYP2C19. Few clinically relevant interactions have been documented for 
the PPls, despite the substantial potential for pharmacological interactions. However, when certain 
medications are co-prescribed with these agents, care should be taken. The frequency of clinically relevant 
drug interactions rises in direct proportion to the patient's age and the number of medications they take.
